The Auto-Mag® Bacterial DNA/RNA Kit utilizes magnetic bead technology for the efficient extraction of high-quality genomic DNA (gDNA), RNA, or total nucleic acids from Gram-positive and Gram-negative bacterial samples. With a single, streamlined protocol and versatile reagents, the kit can process up to 1 mL of bacterial culture per preparation, eliminating the need for organic extraction or bead beating.
Cell lysis is achieved through a specialized buffer and enzyme combination, ensuring minimal gDNA fragmentation. The kit offers flexibility to recover gDNA by treating the lysate with RNase enzymes, isolate RNA with DNase treatment, or co-purify both gDNA and RNA as needed. Following treatment, binding conditions are adjusted, and Auto-Mag® D-3 magnetic beads are added to capture the target nucleic acids. After two rapid wash steps to remove salts and protein contaminants, the purified DNA or RNA is eluted and ready for immediate use in downstream applications, requiring no further purification.
Bacteria DNA/RNA Extraction Workflow
- Prepare bacteria cultures & pellet the cells.
- Add lysis buffers to lysis cells.
- Remove DNA or RNA
- Add beads to bind DNA or RNA.
- Separate the beads from contaminants
- Wash magnetic beads to remove contaminants.
- Elute DNA or RNA or total nucleic acid from magnetic beads.

Figure 1. Genomic DNA or total RNA was isolated from 0.5 ml, 0.4 ml, and 0.3 ml of overnight cultures of Gram-negative E. coli using the Auto-Mag® Bacterial DNA/RNA Kit according to the standard protocol. The extracted DNA and RNA were analyzed separately on an Agilent TapeStation 2200. Figure left demonstrate the analysis of the extracted total RNA, which display high-quality total rRNA with RIN greater than 8.5. The figure right shows the analysis of the extracted gDNA, which display that all gDNA are greater than 50 kb with a DIN of 9.6.
