Auto-Mag® Yeast DNA Miniprep Kit utilizes advanced magnetic bead technology for the rapid and reliable isolation of high-quality total cellular DNA from various yeast species. The kit efficiently processes up to 1.5 mL of log-phase yeast culture (OD600 of 1.0 in YPD medium), making it ideal for streamlined and reproducible DNA extraction workflows.

Auto-Mag® Yeast DNA Miniprep Kit workflow begins by adding lysis buffer to the yeast sample and transferring the mixture to a disruptor tube. The sample is then mixed and heated for 5 minutes. Following this, the sample is homogenized for 5 minutes using an appropriate disruptor to ensure efficient cell lysis. Next, digestion enzymes are added to the sample, which is incubated at 55°C for 20 minutes to release the DNA effectively. The sample is then centrifuged, and the supernatant is transferred to a DNase-free microcentrifuge tube. For RNA-free genomic DNA, an optional RNase treatment can be performed at this stage. DNA is captured using magnetic beads in the presence of a binding buffer. After two quick wash steps to remove salts and protein contaminants, the purified DNA is eluted with an elution buffer. The resulting high-quality DNA is suitable for downstream applications such as PCR, restriction digestion, and hybridization techniques.
