The Dried Blood Spot (DBS) DNA Isolation Kit is a specialized tool designed for extracting DNA from dried blood samples. It is widely used in research and forensic applications, where blood samples are preserved in dried form on substrates like blood cards. This method simplifies the collection, transport, and storage of DNA for later analysis.

The Auto-Mag® Dried Blood Spot DNA Isolation Kit is specifically designed for the efficient purification of high-quality genomic DNA from dried blood spot samples. By utilizing advanced lysis conditions and Auto-Mag® magnetic particle technology, the kit ensures consistent and reliable DNA isolation. The purified DNA is compatible with a wide range of downstream applications, including Southern blot analysis, PCR, and next-generation sequencing (NGS).

For added versatility, the kit provides an alternative DNA isolation protocol using a centrifuge and 1.5 ml tube, making it ideal for situations where magnetic separation devices are unavailable, without compromising performance.

In the Dried Blood Spot DNA Isolation Kit Standard Protocol, Version 2.2, the volumes of the lysis buffer, binding buffer, and MDW buffer have been increased to meet the reagent volume requirements of high-throughput automated purification platforms, such as KingFisher™ and IsoPure instruments. All procedural steps and manual operation requirements remain unchanged from Version 2.1.

Manual Workflow for Dried Blood Spot DNA isolation

1. Cut 3-6 pieces from a dried blood spot and place them in a Nuclease-free 1.5-2.0ml tube
2. Add Proteinase K & lysis buffer to lysis the cells.
3. Add Beads to bind DNA
4. Separate beads from contaminants.
5. Wash magnetic beads to remove contaminants.
6. Elute DNA from magnetic beads.
