The Auto-Mag® FFPE Tissue DNA Isolation Kit utilizes paramagnetic bead-based technology to efficiently isolate DNA from formalin-fixed, paraffin-embedded (FFPE) tissue without the need for xylene. This kit includes specialized deparaffinization reagents and a compatible extraction buffer, allowing the entire process to be conducted in a single tube, reducing sample loss and cross-contamination.

The kit supports both manual and automated workflows in 96-well plates, as well as manual processing in 1.5–2.0 mL tubes. The protocol begins with the solubilization of paraffin from tissue slices, followed by enzymatic lysis to digest the tissue and release nucleic acids. Decrosslinking is performed at high temperatures, after which nucleic acids are immobilized onto Auto-Mag® D-3 beads using a binding solution. Contaminants are removed through a straightforward wash process, and the DNA is eluted using the provided elution buffer. The purified DNA is of high yield and integrity, free from inhibitors, and ready for use in various downstream applications, including qPCR, mutation screening, microarray analysis, sequencing, SNP genotyping, and STR genotyping.

Auto-Mag® FFPE Tissue DNA Isolation Kit is also compatible with automated workflows, including KingFisher™ platforms, the IsoPure Automated Purification System, and most magnetic rod liquid handling systems. FFPE DNA isolation workflow with single tube 

  1. Scrape FFPE tissue from FFPE slides or blokes.
  2. Deparaffinize with FFPE Deparaffinization Reagent (FDR)
  3. Lysis sample and decrosslink 
  4. Add beads for DNA binding
  5. Separate beads from contaminants.
  6. Wash magnetic beads to remove contaminants.
  7. Elute DNA from magnetic beads.
