Auto-Mag® Plasmid DNA Miniprep Kit (Version 3.0) integrates nanomagnetic bead technology, alkaline SDS lysis, and an endotoxin-free plasmid binding buffer to isolate high-quality, low-endotoxin plasmid DNA from up to 1.5 mL of overnight E. coli culture. The purified plasmid DNA is immediately suitable for downstream applications.

The extraction workflow is compatible with a 96-well plate format, enabling parallel processing of up to 96 samples. For high-copy plasmids, typical yields range from approximately 2–5 µg per 1 mL culture sample. Actual yields may vary depending on plasmid copy number, E. coli strain, culture media, and growth conditions. In addition to high-throughput processing, the kit provides protocols for plasmid purification using standard 1.5 mL microcentrifuge tubes, as well as an alternative procedure that does not require magnetic separation instrumentation, offering flexibility across diverse laboratory environments.

Auto-Mag® Plasmid DNA Miniprep Kit is also fully compatible with automated workflows, including KingFisher™ platforms, the IsoPure Automated Purification System, and most magnetic rod liquid handling systems. Plasmid DNA Miniprep Workflow

  1. Prepare bacteria cultures & pellet the cells.
  2. Add lysis buffers to lysis cells.
  3. Separate cell debris
  4. Add beads to supernatant to bind plasmid DNA.
  5. Separate the beads from contaminants
  6. Wash magnetic beads with 70% ethanol to remove contaminants.
  7. Elute plasmid DNA from magnetic beads.

Figure 1. pUC-19 plasmid was purified from 1 mL DH5α cultures harboring the plasmid and eluted in 100 µL volume using kits from Auto-Mag® Plasmid DNA Miniprep Kit and a competitor kit according to manufacturer’s recommended protocols.

Figure Left: Plasmid DNA concentration was determined by optical density measurements using Thermo Scientific’s NanoDrop™ 2000c system. In terms of determined recovery, Auto-Mag® Plasmid DNA Miniprep Kit shows a higher recovery than the competitor’s kit.

Figure Right: Purified Plasmids are cut with BamH1 and, 10 µL from each sample was analyzed on a 1.2% Agarose gel.

Figure 2.  Plasmid DNA purified with Auto-Mag® Plasmid DNA Miniprep Kit was used in a  Sanger sequencing reaction.  DNA was analyzed on an Applied Biosystems 3730XL. Chromatograms show continuous reads close to 1,000 base in length with uniform signal intensity and very low noise or background.
