Auto-Mag® Plasmid DNA Midi Prep Kit is a versatile magnetic bead-based system for efficient, large-scale purification of high-quality plasmid DNA from recombinant bacterial cultures. It can isolate 80–250 μg of high-copy or 10–50 μg of low-copy plasmid DNA from 10–50 mL of overnight culture, ensuring excellent yield and purity. The resulting plasmid DNA is immediately ready for use in a range of downstream applications, including fluorescent DNA sequencing, restriction enzyme digestion, PCR, cloning, transformation, mammalian cell transfection, and more. For added convenience, the kit also offers a non-magnetic separation protocol, enhancing flexibility for different lab workflows and equipment setups.
Plasmid DNA Midi-Prep Workflow
- Prepare bacteria cultures & pellet the cells.
- Add lysis buffers to lysis cells.
- Separate cell debris
- Add beads to supernatant to bind plasmid DNA.
- Separate the beads from contaminants
- Wash magnetic beads with 70% ethanol to remove contaminants.
- Elute plasmid DNA from magnetic beads.
