Auto-Mag® Bacterial DNA/RNA Kit utilizes advanced magnetic bead technology for the reliable and efficient isolation of genomic DNA and/or RNA from both Gram-positive and Gram-negative bacteria. Featuring a streamlined workflow and versatile reagents, cell lysis is achieved using an optimized buffer and enzyme system, ensuring efficient nucleic acid release with minimal genomic DNA fragmentation. The kit offers flexible workflows, allowing users to isolate genomic DNA (via RNase treatment), purify RNA (via DNase treatment), or co-purify both DNA and RNA from the same sample. Following lysis and optional enzymatic treatment, binding conditions are adjusted and Auto-Mag® D-3 magnetic beads are introduced to capture target nucleic acids. After two rapid wash steps to remove salts, proteins, and other contaminants, purified DNA or RNA is eluted and ready for immediate use in downstream applications without additional purification.
Auto-Mag® Bacterial DNA/RNA Kit is also fully compatible with automated workflows, including KingFisher™ platforms, the IsoPure Automated Purification System, and most magnetic rod liquid handling systems. Bacteria DNA/RNA Extraction Workflow

- Prepare bacteria cultures & pellet the cells.
- Add lysis buffers to lysis cells.
- Remove DNA or RNA
- Add beads to bind DNA or RNA.
- Separate the beads from contaminants
- Wash magnetic beads to remove contaminants.
- Elute DNA or RNA or total nucleic acid from magnetic beads.

Figure 1. Genomic DNA or total RNA was isolated from 0.5 ml, 0.4 ml, and 0.3 ml of overnight cultures of Gram-negative E. coli using the Auto-Mag® Bacterial DNA/RNA Kit according to the standard protocol. The extracted DNA and RNA were analyzed separately on an Agilent TapeStation 2200. Figure left demonstrate the analysis of the extracted total RNA, which display high-quality total rRNA with RIN greater than 8.5. The figure right shows the analysis of the extracted gDNA, which display that all gDNA are greater than 50 kb with a DIN of 9.6.
