Auto-Mag® Water DNA/RNA Kit is a novel, patent-pending solution for water sample microbial collection and nucleic acid extraction. Unlike traditional environmental microbial analysis methods, the kit utilizes specially modified magnetic beads to efficiently capture and enrich bacteria, pathogens, viruses, free nucleic acids, and other microorganisms from water samples. Figures 1. Environmental microorganisms’ collection. Add capture beads to 10ml of water sample spiked with 1x10^6 E. coli, mix and rotate at room temperature. Take an equal amount (200ul) of the mixture every 5 minutes and place it on a magnetic device for 2 minutes. Take 50ul of the supernatant and inoculate on the surface of the LB agar plate. Incubate plates at 37℃ for 18 hours and count the number of colonies. Plates (1-5) show that more bacteria are captured as the capture time increases. More than 99% of the bacteria are captured by the beads within 20 minutes. Plate 9 shows that the bacteria captured by the beads can be completely re-cultured.
Genomic DNA Isolation using Auto-Mag® Water DNA/RNA Kit.
Figure 2. Genomic DNA from different volume of lake water sources was isolated with the Auto-Mag® Water DNA/RNA Kit. DNA was extracted in triplicate from water samples as follows: sample 1-3 from 200 ml simples, sample 4-6 from 100 ml simples, sample 7-9 from 50 ml simples, and sample 10-12 from 15 ml simples. Genomic DNA was analyzed on a 1% agarose gel to demonstrate yield and quality of the DNA.

Figure 3. Detection of isolated DNA by Real-Time PCR (SYBR Green). The E. coli carrying an ampicillin resistance plasmid was added to the water sample. E. coli was captured, and DNA was isolated using the Auto-Mag® Water DNA/RNA Kit. The purified DNA was a serial 10-fold dilution for PCR amplification. The results showed that the purified DNA has high purity, and no obvious inhibitors were present.
