Auto-Mag® DTR is consists of AMD Biotech’s own paramagnetic beads and compatible buffers and is designed to remove of unincorporated dye terminators effectively and reliably from Sanger sequencing reaction mixtures. The process consists of three simple steps including bind, wash and elute. While binding the sequencing product selectively to the paramagnetic beads, unincorporated dyes, nucleotides, salts and primers will be removed during ethanol washes. This principle allows for elution of the pure Sanger Sequencing product in the elution buffer of choice. Auto-Mag® DTR can be used for manual procedures as well as guidelines for adapting it to automatic liquid handling workstations (e.g. Dynamic Devices, Beckman, Hamilton, Tecan, Caliper, Perkin Elmer, Agilent and Eppendorf)

Fig 2. pUC-19 plasmid was sequenced using the BigDye Terminator v3.1 Chemistry with recommended protocols. 10 ul Sequencing reaction products was purified by using 10 ul Auto-Mag® DTR. DNA was analyzed on an Applied Biosystems’ 3730XL. The Sequencing reading is over 1200bp. There is no dye blobs and Phred 20 read lengths is over 1000bp
